When my iMac is asleep, once every hour the Superdrive makes a noise like it's checking for a disk. It's the same sound it makes when you wake the computer from sleep but it shouldn't be doing it WHILE it's asleep. It does it once every couple of hours. Not a huge deal...but what could be the issue?
 
Your iMac is probably waking up (the superdrive would attempt to read a disk, making the sound that you hear), then going back to sleep. It's likely a setting on some app that you have installed.
You may find out what is causing the wakeup in your Console, then look at the system.log for the approximate time that you hear the Superdrive reset.

One possibility is to open your System Preferences/Energy Saver, then uncheck "Enable Power Nap".
