if a user is upgrading RAM in new iMac, is there any benefit of pulling the stock from slots 0 & 2 and installing new in those (primary?) locations?

Adding 32GB(2x16) Crucial kit to stock base 8GB(2x4)
 
As mentioned so long as matching DIMMs are in alternating slots it doesn't matter. I just added an extra 32GB of Crucial Ballistix into the empty slots in addition to the 8GB of stock RAM.

Keep in mind that on the first boot after changing RAM configurations there will be up to one minute of black screen while the iMac configures itself for the new memory. Just wait it out.
