If I add a wireless mouse and keyboard to my iMac will this mean that I wont get mightymouse?

Yes, If you choose the Bluetooth KB and mouse as a BTO option you will not recieve the wired KB and mouse.
 
I was at the Apple store last night and looked at the wireless KB. It didn't have any USB ports on it like the wired KB, either config you get 3 USBs
