Not exactly what you asked about, but assuming 2 of the 4 slots can be empty , this should work.
Of course, you'd want to run all four channels for optimum performance.

Retailers are the ones who heavily promote that to get you to buy more memory.
Most users aren't going to notice the difference between 2-4 DIMMs where capacity isn't a factor, the difference between 3 and 4 is tiny for content creation workstation performance.
Buy DIMMs from the same manufacturer with the same specifications you'll be fine. Filling all 4 channels shouldn't be something to try attain, just go for the capacity you want.
I see… I'm going for 32GB and I don't see a way of doing that affordably without filling all slots.
Those 16GB DIMMs at Superbiiz just dropped massively in price. $340 for two and shipping. That's what I'd go for if I were you.
