Long story short, every beta up until now has been a breeze and gone without a hitch. However, having just returned from holiday (didn't take MacBook with me), my App Store simply refuses to acknowledge that beta 6 is even a thing. Anyone able to help at all? :(

Try the terminal command:

sudo softwareupdate -i -a

This will update the system using terminal.

There's an issue with DB6 in that it installs but then doesn't show as having been installed in the App Store.

My App Store shows DB5 as the last beta I've installed, but I'm running version 15A244d which is DB6.

Might be worth checking, just to make sure you really don't have it already before wasting time trying to find it :)

Yes, you might already have installed and forgot about it.

I installed the update of DB6 on several Macs, some with a clean install starting at DB1 and others that where already on DB5. On none of them it was shown as installed after that.
