I don't know if this will be relevant to you at all, but I have problems with the eSATA on that card. My G-SAFE (Raid 1 hard drive) drops off every once in a while, meaning the Mac Pro will not recognize it and sometimes will mess with booting, disabling my keyboard.
That's a bummer. I only have two remaining eSATA drives left, both older LaCie externals, and they still work great with eSATA to the CalDigit card. They're those kind with FW400, FW800, USB 2.0 and eSATA, so if the eSATA were to give me any trouble, I'd have to revert to the slower FW800.
I got this card specifically because it doesn't require a reboot to see newly attached drives, like my older, cheap, $50 2-port eSATA card did. Every now and then, a drive will hesitate to show up on the desktop, but a quick power off and back up on the external drive brings it up. It's so much faster via eSATA that it's worth flipping the switch instead of going FW800.

Did you look on the manufactures website? They list the specifications there along with the supported operating systems.
Received mine today.
Previously had 2 different cards, the Caldigit USB3 plus a Stardom SJ-20 eSata.
The stardom has a fan & was quite noisy.
Now all working well with the Caldigit FASTA-6GU3, & quiet!

Does this Caldigit card require to connect a power cable to the motherboard?

No, it doesn't use any power cables.
I can't answer about sleep, because I don't sleep my system... I just shut it down when I don't need it. (The SSD boots the system so fast, it makes sleep seem pointless, and I don't need to experiment with conflicts with my RAID.)
The CalDigit FASTA-6GU3 card installed in slot 3 or 4 in a Mac Pro 3,1 (2008) will only be in the PCIe ver. 1.0 slot. How will that affect USB3 performance?

As I understand it, the card is x4 lane, but utilizes a PLX chip / switch that is only x1 lane. I've used all four ports on the card at once (2 USB3 and 2 eSATA) while doing data transfers, and it didn't seem to slow down, but I'll admit I wasn't benchmarking at the time. Perhaps each port gets a lane in such a case, or perhaps not, but I've personally only seen as much as 246MB/second when specifically speed testing an SSD that runs at 456MB/sec when connected to my Areca RAID card directly.
Is the max 250MB/second on a single port? Maybe, but it does work perfectly, and is hot-pluggable. I've not personally tried a RAID of multiple ports at the same time, which would be interesting. Given PCI 1.0 is 250MB/sec per lane max, it may make no difference in slots 3 or 4 on a 3,1 Mac Pro.

Cal digit fasts-6gu3 sleep issue
I just installed one and I got the same not ejected properly message with a USB 3 drive attached. Other than that it works great. It's like having new external hard drives.
When I put my computer to sleep and there is an USB 3 stick in the card, I get a message that the stick is not ejected properly.
I hope this will be solved in an update.

I installed the CalDigit card, and attached to it a SanDisk USB3 card reader. All works, and it sees my SanDisk Extreme Pro cards, SDXCs and CompactFlashs.
However, it feels really slow.
When I tested the two types of cards with AJA, I got satisfying results, writes around 85MB/s and reads around 90MB/3, which is acceptable as the cards are "only" rated at 95-100MB/s. But when I am actually transferring Nikon D800 NEF images from the CompactFlash card to my Mac Pro it is much much slower.
Does the type of files make a difference?
