I have an amplifier that can be driven by a USB connection from my MBP, but I would still like to have sound sent to my speaker-out jack on the computer itself. When I use the external amp with the USB, I have to change the sound drivers on my computer, and the speaker-out jack is silenced.
I know this is a long shot, but is there any program/technique/setting that will enable both devices to play the same audio simultaneously?

i use an airport express and the audio jack.

the airport express plays itunes to my sound system.
the audio jack plays system sounds.

that way guests listen to music, and anything i am doing on the computer doesn't come over the stereo. it gives me a good point of reference to what's going on.

however, the express ONLY plays itunes. it will not play any other media (at least that i have been able to determine). you can also tell itunes to play on all outputs so it will play through the speaker jack AND the express both at the same time.

i hope this helps, but i don't think it's what you were asking for.

I need to use both sound systems at simultaneously playing the same audio source (I'm running sound for a small event and need to set it up like this:
MBP > USB > amplifier > stereo speakers (which lack a subwoofer); and
MBP > stereo out > computer speakers w/subwoofer.
w/both speaker sets playing the same audio source.

how about playing both of them through the audio jack?

use one of those little splitters that allow 2 people to use headphones off the same jack to listen to one ipod. (3.5mm male to two 3.5mm female - available at radio shack, walmart, etc.)

or is that solution too simple?
