In order to connect an eGPU to an iMac with and external display one would connect the eGPU to the iMac and the external display to the eGPU correct? Then set the external display as the primary display via system preferences?

I just want to make sure this is feasible as I am leaning towards getting the 27 inch iMac (i5 3.0 ghz, Radeon pro 570x, 256 gb SSD). It is much more economical at this point and if I decide I want to do light gaming I can add the stated eGPU set up at a later date. I just wanted to make sure that is feasible?

Yes

For some apps you don’t need to set “prefer external gpu.” At least in FCP - it’s smart enough to optimize on its own.
