Browser (Safari, Firefox, Opera) > Preferences, if you mean such downloads.
For other applications, the download folder should also be modifiable via the Preferences.

Does anyone know how to get the downloads to go straight to the desk top instead of the download folder?
You can change this behaviour in Safari's preferences in the general Tab.
