Could I create a folder somewhere and in it have a Numbers spreadsheet, a photo, an Apple Note, an audio file, a video, a .PSD file, etc.?

If so, what happens when you click on a file, does it ask you which app you want the file to be opened with?

Yes, you can put a mix of files into a folder. When you tap on the file, it’ll either open it up in the app you created it in (such as keynote) or if it’s a PDF or image, it opens it up in a preview and then you can do what you want with it via sharesheet.

Can you Air Drop files from a Mac directly into a designated folder? OR when Air Dropping, does it just go into the Files folder to be sorted later?

Is there a way to access the Files app on a Mac directly?
