I'm now running two video cards:
GT120 (for boot)
and (PC) GT640

Once logged in, is it possible to make the GT640 the primary screen (without switching the cables around)?

Displays prefpane, drag the menubar to the appropriate screen. Assuming you're not wanting the 640 to power a display that's plugged into the 120?
 
The SysPref-Display is very interactive. You can drag the screens up or down and switch positions. And as noted above, move the menubar to the "main" display.
