How to reformat portable external hard drive?

My WD 2TB My Passport was replaced and I cannot find the guide I used to reformat it with Disc Utility before. All I can recall is it had a partition with 250GB formatted to backup my Mac, and the remainder I believe was formatted differently to store my files. But I cannot remember exactly how it was done and want it done right the first time.

Does anyone know of such a guide? Or could someone provide me the steps to do this?
