I have an AT&T HTC One. My bootloader is unlocked.

When the HTC One Nexus RUU becomes available, will I be able to flash it and then from there-on, receive updates the same time as anyone who has an HTC One from the Google Play store?

probably NO.

Reason is your CID (model identifier) will still be reading as an AT&T HTC One and therefore when it checks for updates it uses that piece of info to retrieve update information and of course in this instance it simply wont find any new update because the CID and the Rom are two different things.

You will likely have to S-OFF your device which should be easy now you are bootloader unlocked.

You can then change you CID to SUPERCID 1111111 which should allow you to recieve the OTA update or alternatively change the CID to the same CID as the Google Edition, effectively fully changing your device into an unbranded google edition model in every way.

Unlocking the bootloader on the HTC One is extremely easy. HTC allows it.

While I agree Samsung phones are easier to root and ROM, HTC has made it easier to hack their phones. I'm posting this from my AT&T One while running the Google Edition ROM from XDA. I'm not S-Off yet and haven't changed my CID. Just unlock the bootloader through HTC Dev, root, and you're good to go. Now, if you want the updates to the GE ROM straight from HTC, yeah, you're gonna need to change to SuperCID. But with the source out there, you can just use the XDA GE ROM and update it when the new sources are out.
