I bought iPhone 4 two months back it was 4.3.5 version , it was working fine but yesterday i upgrade this with version Version: 5.0.1; but now it got locked every thing is working except sim. I am really worried , I don't know what to do ...Please help me somebody..............

Anyway, I bet the iPhone was jailbroken and unlocked when you bought it. And now that you've updated it, it is locked again. Try putting in an AT&T sim card in the iPhone. If you call from it, the iPhone is locked to AT&T. If you're lucky, you can get AT&T to unlock it for you.
