Last night I had connected my HDTV's HDMI to my iMac 5K through MiniDisplay port, and it worked!

Then I unplugged the HDMI to check something, and plugged it , but iMac couldn't detect it! I tried many times but it couldn't again (even in both ports)!

However, I plugged it into my late 2008 MacBook and it has detected it!

Is my iMac's thunderbolt has became suddenly defective, or something wrong went with MiniDisplay port?

It may sound silly but have you tried restarting your machine?

I am asking since it sounds very unlikely that both ports have stopped working at the same time so it could just be sure software glitch.

Yes, I did restart the machine and tried various solutions!
 
What else have you tried? That might help me and others get an idea of what it might be. Also, is both Thunderbolt ports recognized in the System Report?

I've only tried many simple restart and turn off solutions! I've just read about SMC and PRAM reset, I'll try them later after work!

I don't think that the Thunderbolt port gone defective!

How could I double check the Thunderbolt ports in the System Report?

What Samuelsan2001 said is probably a better solution. But if you want to find it you go into About This Mac -> System Report and then in the left hand side there is a tab called Thunderbolt.
 
I checked the System Information, it seems that the port is fine, a screen photo is attached!

Keep in mind that the Mini Display port was unplugged during the report!

It worked!

I conducted a PRAM reset and it worked!
