When switched on recently my iMac (2010) loaded itself slowly into safe mode. The keyboard definitely wasn't pressed. It was quite slow to load too, with a grey loading bar below the Apple logo. Since that point it has booted fine (at least 3 times). What could have caused this and could there be something wrong? Thank you for any responses.

While it is curious the Mac did this on its own without prompting from you I don't think you have anything to be concerned about. If it did this continuously you probably would want to disconnect/switch out the keyboard just to make sure it wasn't getting input when it shouldn't be. If it still happened after that, you'd want to proceed by resetting PRAM. This would clear any commands inputted to run at startup in the firmware. Incidentally, you could tell the computer to always boot in verbose mode if you so desired.
