Just purchased a refurb iMac 2.93ghz, noticed dust under the screen, well I think its dust as I can still see it when the mac is turned off, would a dead pixel be visible when turned off?

Is there anyway I can remove it without taking the glass out, maybe compressed air?

A dead pixel would not be visible ever, its dead. You can remove the glass with suction cups. There are guides for this on the net, google "iMac hard drive replacement".
 
Its dust the pixels arent active when the computer is off. The screen isnt airtight so the compressed air might work, but keep in mind that dust will just go into some other part of the computer and may or may not cause damage
