Hi

I am not a programmer. I just wanted to know if something is possible.

Is it possible to interface with the in-built phone application? I'd like to be able to make an application that can answer the call and turn off the ringer and play voicemail to the caller like an answer machine would.

No. The SDK does not give any way to interact with phonecalls at all. This may be possible on jailbroken phones, I've no idea about that.
 
Reason I ask is I saw an app to disguise voice in Cydia and also to fake the calling number so I thought some interaction must be possible.

Cydia == Jailbroken phones. It's not possible using the official SDK so any such app is not possible for the App Store. These forums tend to be frequented by programmers using the official SDK only: if you want help with apps for jailbroken phones you'll probably be better off elsewhere.
