I'm about to smash this iPhone, after I updated to 9.3.4 the battery dies completely after 20 minutes of being charged overnight.

Only thing I can think that went wrong... I used BatteryDoctor while the phone was inside a mophie, after that, the battery hasn't been the same.

1- battery drops from 100% to 80% within 5 minutes on instragram.
2- Phone dies at 60% battery life

Anyone else having this issue?

Perhaps try updating to iOS 9.3.5 and see if that makes a difference?

Make sure all apps are closed out. Do a hard reset then do a reset all settings.

I'm not familiar with Battery Doctor, but I use an app called "Battery Life" and it will tell you how much wear is on the battery. You could use that to see if your battery is still good or needs to be replaced. If it's still good, then try what wymi said (close all apps, hard reboot) and if that doesn't work, try recovering from an old backup or do a full factory reset.

I did a hard reset, and reset all, also I updated to the latest iOS, and still have the same problem.

Battery Life is telling me I need to replace my battery, I guess I have to trust that app.

I wouldn't take a 3rd party app's advice yet. I would try a clean install first. Plug it into iTunes, have it download the latest iOS and reinstall. Manually add back your data.

If you're still seeing bad drainage like this, then you probably have a hardware issue, i.e. a bad battery.

Considering its an iPhone 6, its very likely its a hardware problem and not software. Especially that rapid of a drain and shutting off about 50%. The 3rd-party app 'Battery Life' is extremely accurate when it comes to battery life health/remaining capacity. I have compared it to Apple's own diagnostic on a few devices and its within 1-2%. The app is reading the battery directly.

Interesting, I didn't think they were that good. I was trying to save the OP from a potential unnecessary trip to the Apple Store, but after hearing this, the trip should be made.

Apple will run a test on the battery before replacing anyway. But the OP can also call and they will run the test OTA to confirm.
