Hi guys... is the iMac update a proper update? Or is there likely to be a huge one in the next 6 months with a completely different design, tech specs etc?
 
It is the update of the year for the current iMac. If a redesign happens, it will be in October or most likely in 2012.
If you need it now, wait. Or something like this.

Also is it best to buy the best Hardrive which is, 2TB Serial ATA Drive + 256GB Solid State Drive

The iMac has gone thru its yearly upgrade (Pretty big jump from Dual-Core/Old i5/i7 to All Quad-Core/2nd Generation i5/i7). Id get whichever Hard Drive you think will best suit your needs, if you need an SSD, get it from the factory as its easier than adding it later, but as far as internal storage goes, you have Thunderbolt, so adding fast external storage shouldnt be an issue.

We don't know for sure, but Apple seems to adopt a yearly update cycle, especially since new CPU microarchitectures (Sandy Bridge and the like) only come out once a year and any update will be just a speed bump. Maybe Apple surprises us all with a Fall update and redesign, but we don't know. It is all just speculation, thus my words should be taken with a cup of salt.

That depends on your usage patterns and how much data you want to store inside the iMac. Know that TB enclosures for HDDs and SSDs will be available soon (June), thus negating internal HDDs almost completely.

An SSD is loads faster than a normal HDD, thus you have a faster computer. The 256GB SSD will house the OS and applications you install, the HDD can be used for whatever purpose you see fit.
