2010 Quad-Core 2.8Ghz Mac Pro
OSX 10.8.4
24GB RAM
4x2TB Internal drives
HighPoint RocketRaid eSata PCIe Card
SanDigital 4-bay JBOD eSata Enclosure 4x2TB
-> LaCie PCIe USB3.0 Card
-> LaCie Porsche Design P’9230 USB3.0 Drive 750GB (used for OS TM backups)

First Issue:
The USB drive will not mount unless I restart the computer, it does not work as "Plug-n-Play".
I dont have another USB3.0 computer but I tested it on my USB2.0 MBP, it mounts perfectly fine on that computer no restart needed.

2nd Issue:
My computer runs like crap when the drive it mounted.
My mouse sticks and freezes and the system just lags.
I dont see any processes killing CPU or memory usage.
Once I unmount and turn off the drive, the computer is back to normal.

- Only LaCie drives work with their PCIe card, which is lame but thats besides the point.
- I spoke to LaCie about the issue and they suggest I take the computer/drive to a repair store to have it checked out? WTF???
- The eSata stuff works flawlessly, however I have not removed the eSata card to see if it's interfering with the USB3.0 card. I may try that tomorrow...

Any help is appreciated!

I had a frustrating experience with the card too. It actually burned out on me. I replaced it with a caldigit fasta card and have no problems with usb 3.0 or esata.
 
I also have the CalDigit USB3 card, the version without the eSata ports, and it has worked well for me. No issues to report. It was recommended by a fellow MacRumors member. I have mine connected to a Drobo 5D and although it is not as fast as TB, it gives me enough speed, ~130MB/S, to run my iTunes media server along with several VM's.
