I was wondering if I would run into any issues running an Asus 780GTX in my Mac Pro with 2x2.4 GHz 6 Core (E5645) 64GB RAM? I have the latest macOS. Do I also need to buy an 8pin? The card takes an 8pin and a 6pin, not sure what my mac tower has because I'm not at home.

Yes, powering a 780 can be tricky, your cMP only has dual mini 6 pin aux power source, but it can be done.

Depends on the real power draw, you may only connect them to the mini 6pins (780 should be OK), or use an extra SATA port to supply enough power (if drawing too much from the mini 6pin, a self shutdown protection may kick in).

Also, AFAIK, there are 2 revision of the 780. Rev A is supported in MacOS OOTB, but Rev B require web driver, otherwise, black screen only. However, I am not 100% sure on this matter.
 
I ran my classified 780 with mini six to eight pins without issue so I imagine you would be fine.

In terms of rev A vs B, wasn't that for the 780ti?

Edit: You are correct, GK110 B revision issue also affects 780

I ran my GTX 780, Gigabyte 3 fan version with an MVC EFI quite awhile before sawitching to the GTX 1080. It has two 8 pin connectors. MVC shorted out two of the pins on each connector and I ran with the two 6 pin cables. I never had any issues with it. I also have a cMP 5,1.
