Mac Pro won't boot after OS X 10.10.3 update

Firmware from 4.1 to 5.1
Xeon W3680
48 GB 1333 Mhz
GTX 680 Mac Edition
Wireless ac + bluetooth 4.0

Everything was working perfectly until I tried installing the 10.10.3 update, now every time I try to boot I'll get the prohibitory sign. I tried disconnecting all peripherals, safe boot, disk repair, permission repair, SMC and NVRAM reset, time machine restore to 10.10.2, but nothing seems to help.
Windows via Bootcamp works normally, so it must be a software problem.
Do I have to reinstall OS X?

Though the thread starter was using a different Nvidia card. He resolved the issue by getting an older Mac Radeon 5770 OEM GPU.

Shouldn't be the case, my card is a Mac Edition, it should work with default OS X drivers...

Yeah. You're right. I was also wondering and puzzling why the update causes problems. Sorry I could not be of much help. Before installing new OSX updates, I usually make sure I have a clone of my original OS so it's easier to revert back.

Just read the trim enabler support page, I forgot to disable it before installing the update. Thank you for the help!

I've been brought there several times. The worst case was a mix between trim enabler and nvidia web drivers and ended up reinstalling OS X on top of my installation to recover.

Yes, and when TRIM Enabler disables KEXT signing, it sets the Nvidia driver to native OS X driver.

Unfavorable if you need the Nvidia web driver for newest GTX 9xx graphics card….

But the OP has a compatible GTX 680 Mac Edition, should be no problem to disable KEXT signing, and after a restart setting Nvidia web drivers again, and then of course a second restart.

Yes, Cindori acknowledged the sloppy code work and may some day fix this. His Trim Enabler assumes that you don't have or need any other boot args and overwrites/deletes anything else there. The Nvidia Web Drivers are smarter than this and just ADD their arg in. So if you are running both, install Trim Enabler first, then the Web Driver.

AT least until Cindori fixes his code.
