To use some thing like Macrium Reflect Free Edition do you have to boot to it? You suppose to make bootble USB stick with Macrium Reflect Free Edition?

If you're talking about backing up a Boot Camp partition, while yes - Reflect (Acronis, etc) will take a backup of your partition, it's not necessarily going to be usable immediately if you happen to lose your BC partition - especially if you're asking really basic questions like the above.
