I've got an iPhone 7 Plus and My mail app uses 5,38 GB of Storage. I deleted all mail accounts, rebooted the phone. Readded my accounts but still it shows up 5,38GB of storage used in my phone! latest iOS 10 Version running! I don't want to restore my phone. I synced it also with iTunes but nothing worked out! Can anyone help??

It's very possible. Mail is caching all of your mail and attachments onto your device. Go into settings and mail section and into each account and see how many "mail days to sync" back the email accounts are syncing. If set as no limit then it is every email for every account going as far back as you had the account.

If you have a few mail accounts its very possible to take up a few gigs of space on your phone for thousands of tens of thousands of emails and attachments.
