In mail. Right Click on the Inbox folder. Select Account Info.
If you have multiple email accounts, there is an account dropdown box, select the various accounts and it will summarize the number of messages and size of each mail folder for the account.
Yes, TM does backup the mail folders, including Inbox. If you ever have to clean install and restore, or migrate to a new Mac, everything will be there.
