Hi, i use Maps to find routes to a location. I have ‘Avoid‘ enabled next to Tolls and Highways but the app keeps giving me three choices:

Option1 Avoid tolls: A faster route is available with tolls and highways
Option2 Avoid tolls: A faster route is available with tolls. Highways required.
Option3 Fastest route: Route required. Highways required.

How can I make it displays routes that do not require tolls nor highways?

It’s telling you the highway is required though and I assume if you knew the route better you wouldn’t need navigation

Use Google maps and compare the routing. If the highway is still there, it might be required to get to your destination within a reasonable amount of time.

In this case, Maps presented three suggested routes. Can we increase the number of suggestions?

At that time I tried Google maps but as I recall, it suggested the same three routes. At least what appeared to be.

In Apple Maps, can we also set the number of minutes as the threshold for 'reasonable amount of time'?

No. And unfortunately they only have the “avoid” options. It would be nice to have “do not route on” options. That way all suggested routes would be closer to you desire.

If this is important to you, professional GPS apps provide means to designate any route you want. For instance Copilot has an option to tap a street and it will automatically route through there. You can tap to shape any route you want, or just once to have it choose the best way to get through the point. I used it professionally for over a year all over the country and liked it. It’s not cheap though and in many respects Apple Maps is better.

Some destinations have highways and tolls required and no back roads are available. Use another app like Waze to check if that's the case.
