I agree with a lot of other people that this is supposed to be a secondary laptop for most people. I'm sure it will be Paris Hilton's only laptop, but for those of us who work and travel a lot it will be a secondary. The only problem is the lack of Target Disk mode. To be a useful secondary laptop it would work best to keep your email files, and other needed files, on the MBA's home directory. You would then connect it to your desktop via firewire as an external hard drive and using aliases, be able to use your email and files. Then when you are ready to go on the road you just grab your MBA and you have all of your files already there. There would be no need to sync.
Now, I can place the MBA next to my desktop Mac and connect wirelessly to it, but let's not pretend this will really work. First, it will have to be through AFP, secondly it will be much, much slower than a firwire or USB2 connection, and secondly there will be all of the authorization issues and permission issues to deal with.
I don't object to the lack of a firewire connection. I do object to the fact that Apple has not designed this to become a USB2 target disk. Maybe it will be an update, hopefully software and not hardware.
