Followed the steps on the forum and watched a bunch of YouTube videos, I flashed my RX6600 on PC and it won't boot up on my MP5,1 (running Monterey),

I put it back on my PC, but it won't boot either, what to do to reflash it, at least make it work on my PC..🤣🤣

You need to have it installed with another GFX card known working in your PC and flash it back with the original bios you should have backed up before flashing the card.

So, I will need a motherboard that supports dual PCIE, right?

I tried and it did not work,
the PC shows 2 graphic cards in the device manager but can get the amdvbflash run, saying "Error reading from ROM" (driver installed)

You may need to force flash a bricked card in order to recover it. DOS prompt (as admin) usually works better. In fact, if possible, use Linux.

If that still doesn't work, you may need to short some pins before you use amdvbflash.

And your last resort is to buy a hardware flasher to flash your 6600.
