I'm planning on buying an MX300 Crucial 1TB SSD for my iMac (that has a HDD in it) and then buy an external 2.5" USB 3.0 enclosure. Will this work for running Mac OS off and having it as the boot drive?

Nope, any USB>SATA dock/enclosure would work fine.
Hold Alt on startup when it's plugged in. Hold CTRL when you have the volume list visible & select the external disk with the arrow keys. The arrow underneath the volume will change icon. Then press Enter. This will boot into it & set it as your default boot drive.
So this would work? I just want it to boot without holding ALT every time.

Even better Silicon Power makes a 240GB Thunderbolt SSD which requires no caddy or case. Just select it as the boot drive in System Preferences. If you keep an OS on the internal be aware Software Updates will still go to the internal and a second update to the Thunderbolt.

Just set the USB3 SSD to be your startup drive using the Startup Disk pref pane.
