Does Password Autofill works with only one 3rd-party at a time?
I have 1Password and Enpass installed but can select only one of them along with Keychain in the Settings

Yes only one third party is allowed alongside Keychain at the moment

Probably but I don't think anyone outside Apple could tell you the reason. Maybe submit a feedback report and suggest it to them?
