My PC recently died and am still working on getting it fixed, in the mean time I haven't been able to sync any of my contacts or anything. So I decided to go for the MobileMe trial(will most likely end up purchasing it).

So now my question is, simply for backup purposes, will I be able to enable sync with MobileMe and it send all my contacts to MobileMe and not erase them from my phone?

Or does it work the other way around?

You will need to get the Contacts off the iPhone first. Not sure of the options but maybe you could do it with iTunes while directly connected to the iPhone. I originally used the Yahoo Address Book (iTunes can Sync you iPhone with it) prior to MobileMe. I now use Outlook Contacts. Once you get your Contacts into something that you can upload to MobileMe (via MobileMe Preferences) then when you install MobileMe on your iPhone it will ask to replace your iPhone Contacts (and Calendar) with whatever is on MobileMe.

Sadly I don't think it's possible to get the contacts off my iPhone through iTunes without syncing which will want to sync the library of the computer I'm on with my iPhone, which will wipe everything off of it. If there's at least a way to get all my contacts off my iPhone then put them on MobileMe via Outlook/iMail on another Mac/PC that'd be wonderful.

I recommend you search Google for other Sync Options. Google itself has a way of Syncing with Google Address Book. Just not sure if you can first go from the iPhone to Google. Again, there are other options.
