Say I rip a DVD that contains 4 episodes of a TV show using RipIt and it outputs it as 1 file. Does Plex recognize each episode? Thanks

I really really doubt it. You should rip the DVD so that it outputs 4 separate files, one for each episode. Then you name the episode accordingly (Totally Awesome TV Show - S01E0x).

I agree, Plex reads each of the file names as S01E01 for example and goes to locate the metadata. If you only have one file it will only read it as one file

Ripit is just dumping the DVD, I'm not even sure if Plex can read DVD images.

Use MakeMKV instead, it will dump each title to a different file. Almost all TV shows use separate titles for a new episode, though there are a few much use 1 title with multiple chapters :(
