is there a way to determine the runtime of a Phone? I have a IPhone SE with iOS 10 and want to know how much time has gone since the last time I started it.

Take a look at the battery stats at the bottom of the Battery screen in Settings. It wouldn't really be since the last restart as much as since the last time you fully charged the phone.
