Trying to upgrade the blade SSD with Samsung 1TB m.2 2280 type SSD with the Apple connector adaptor, however, heard that the M.2 typed ssd needs to be setup first (with enclosure) before physically installing it in iMac, otherwise, it won't be recognized at all. So, just checking is this really necessary? If it is the case, is there any particular reason why it won't be recognized initially?

I believe you need an adapter, NOT "an enclosure" (for internal installation).

BE AWARE that using some 3rd-party drives/adapters may "block" firmware updates and software updates in the future. This is a problem with some Mac laptops, may affect the iMac line as well.

Yes, adaptor will be needed when installing the SSD on logic board, but what I was asking is if pre-setup needed before this installation? Which is to install Mac OS, like Catalina into this SSD first?
