Slideshow Play/Pause Button disappeared!

It's there.

You can only see it when you're viewing the thumbnails of a picture.

according to that, you should be able to press play within an individual photo.

Definitely AWOL

I thought I was imagining it but you're right, it's gone. The original button was there in photo view, not just thumbnail view (see below), in fact that is the image still shown (incorrectly) on the iPhone pages at:

http://www.apple.com/uk/iphone/iphone-3gs/photos.html

So maybe it is just an Apple gremlin that crept in. There is also a slightly more irate discussion of this issue on the Apple support forums.
