After old hakintosh ssd broke , I have installed into a new SSD using old opencore hakintosh big sur usb but I am not able boot without usb plugged in .With usb plugged in , everything is working fine .
As expected in this situation , my Gigabyte H410 M is not showing boot drive in boot option but I could see in Sata option.
I have taken a few images for your kind perusal and I am posting them.

CSM , Software Guard , Super IO , SecureBoot => disabled
tried by changing from iMac20,1 to iMac19,1 in config.plist by GenSMBIOS-master then applied ProperTree { oc snapshoot, oc clean and save } .
Please , help.

I assume you've set your BIOS to startup from the Big Sur drive and the OpenCore EFI on that disk.

Have you followed all the post-install directions at the OpenCore install guide?

Once you get this resolved be sure to copy/backup the EFI folder that's on the startup disk's EFI partition to a safe place. I'd think that if you'd done that the first time you could have just re-installed Big Sur with the USB and then copied the backup of the EFI folder to the EFI partition.

Yes , I have gone through again all manual and followed them so far I could understand . I just "erased" my boot drive to APFS with GUID and re-install OpenCore macOS Big Sur . And this time too , I followed all thing mentioned in

Grabbing OpenCore off the USB https://dortania.github.io/OpenCore-Post-Install/universal/oc2hdd.html#grabbing-opencore-off-the-usb

still I have the same problem . BIOS's Boot Option is not showing my OpenCore macOS boot drive so naturally I could not boot without the USB . I am writing this from there .I have noticed that I could not change "Serial Number" shown in "About This Mac" though I have applied GenSMBIOS-master all operations including "Generate SMBIOS".And I have nothing to multi boot as I have booting to macOS nothing else.

I think that you may have more than 1 partition (not including EFI boot partition), right? If so, you may have multiple EFI partitions and your computer is booting a wrong EFI partition, I think.

Yes I had not now . I have deleted and formatted to single partition GUID APFS .
After my previous post I have done exactly that but no luck.
Obviously BIOS is not seeing boot drive in boot option.
Last time and also my 1st time of successful installation of open core macOS sig Sur , I had this exact problem then I got through by changing a boolean value in config plist which is not suggested . But in my case that particular opposite value change works.

If I had done anything else along with that I could not remember as I might not be fully aware of .

Only Clover Configurator is giving me destination EFI / disk1s1 not MountEFI so I used that to place EFI from USB to disk1s1 after my 2nd fresh installation without any partition.I still have the same problem .

The case is solved .
I was coping Boot and OC where I have to copy whole EFI .
When I am mounting the boot drive's EFI , it is mounting EFI so I thought EFI is there and I copied Boot and OC that is a mistake .
