Good question, given that Snow Leopard was mostly under the hood changes. I'd go out on a limb and say any under the hood changes to Lion will be minimal. I'm sure some new APIs are there, but I'm not so sure about quicker book, or responsive.
I think the back-end changes are no-brainers. As far as API's go, yes - I expect there to be a lot with full screen apps and Mission Control. I also expect Apple to provide developers with new API's for things like LaunchPad and tighter system integration.
Those are probably what the Mac only users want.
From the side of a PC network administrator or computer security techie, I would hope Lion could talk well on a PC network, or at least as well as OS X Tiger did a few years ago. Apple has paid so much attention to iPhone, iPod, iTunes, and iPad in recent years, that "networking" a Mac to a PC network has fallen off Apple's to do list.
I don't see Apple ever entering the larger LAN and WAN operating systems market anytime soon, so it's important for those in business that if there is a Mac workstation, that Lion could work as seamlessly as Windows 7 workstations on a Windows server network.
This may seem a small point for many who are home users, but Apple has really put all their energy into their great devices and have paid a lot less attention to OS X. After Lion, which I hope is the last "cat" in Apple's line, it may perhaps be a good time to move on to an OS 11 with a completely redesigned interface.

On the contrary. Lion adds a shitbunch of new APIs, as has every release before it. It's just that Snow Leopard focussed more on that than user-visible changes.
