I might need to exchange a Note4 for another Note4. Is there an easy way to transfer everything from one Note4 to a new one, including home screen layouts, folders, etc. I'd prefer not to root.

Nope. The feature exists in the Moto X and will be available in Android 5.0 Lollipop. But for you right now, you would require root and CWM/TiBu.

Try the smart switch app from samsung. It transfers most of everything except apps including launcher settings (only touchwiz).

For apps transfer you can try helium.

YOu can use the Kies as other suggested. It should cover most of your settings and information. For screen layouts and things of that sort, what launcher are you using? most launcher offer backup/restore option.

The only thing I've never been able to preserve when transferring Android phones is the screen layout and game progress. All my music and pictures are saved to my SD card. My numbers, messages, wallpapers etc are synced to my Samsung and Google Accounts. The only thing I have to do when setting up a new phone is rearrange my home screens.

This. Use the manufacturer backup/restore solution and the backup/restore integrated integrated in to your launcher. I'm not sure about Samsung, but LG allows for an app/settings/password backup to the external sd card as does Nova Launcher.
