well, first you want to take a look here to better understand the facebook API and how facebook apps are structured and implemented.

Flash is not a requirement for facebook apps. However, Facebook and Adobe did release an "official" Actionscript 3 API for Facebook last week (March 31st 2009).

has the API and some diagrams showing some flash/faceboook server architectures.

As for flash versions, you are looking at FP9 and above, as the api is AS3 only.

As for how many application servers you will need? That I can't answer. It depends on your app and its popularity.
