Why would someone prefer an iPhone 6S+ over a 6S? What is there to gain as far as abilities and potential?
 
Pretty much what others have said. I much prefer the larger screen and a bigger battery vs. a small screen and smaller battery.

How much larger is it?
Battery wise, it is not that large, or not so large that justify its significant price rise.
Hardware feature wise, just take an example: optics image stabilisation, now both for video and picture, in iPhone 6s Plus.
Other software tweaks like better landscape on screen keyboard, iPad-like settings app, mail app etc. uh, wait, something is missing?
Yes, those pluses are not that enough to justify "plus" and the price. Maybe as its name suggests, a "plus" model, plus something only suitable on larger phone size.

Why would one have the Plus instead of an iPad?
Different purposes.

I have the iPad Air 2 and an iPhone 6s+. I also have several Macs at home. Every one has a specific purpose.

I don't care to have my phone acting as a GPS Nav system in my pre in dash Nav system car (1997 Honda Accord). I use my iPad for that. I use my iPad for media uses and browsing websites in bed. I use my Macs for email and graphic design with web browsing.

My 6s+ I use as a phone.

Reading for any duration used to be a bit of a joke on the iphone 5 and 4s. I got the plus to start reading and boy oh boy, it's one of my favorite devices for reading. That is one major reason I'll never go back to a smaller screen.
 
It be interesting to use 6Splus as a Nav for the car though wouldn't it?
 
It depends on several factors such as needs, lifestyle, concerns with vision, or preference in usability. The plus does have perks over the 4.7 inch model, like OIS, better battery, more immersive, but some people will still choose the smaller iPhones due to usability, portability, etc.

Side Note: Rumor has it the 4.7' iPhone 7 will have a bigger battery, potentially giving the device better life. We'll have to wait and see. Hopefully, it lasts as long or longer than the improved battery life of the SE model.
