Sorry if this seems an obvious question, but I have an 8TB HDD Time Machine formatted to HFS+ and am now adding two internal NVMe SSD's in my MacPro 5,1 that will store video & audio material.

Should the SSD's be formatted as APFS and if so, will my HFS+ Time Machine have any difficulty accessing/backing up those SSD's?

No, it wasn't until I think Catalina or Big Sur that Time Machine allowed APFS for TM volumes (and completely changed the mechanism by which TM works), up until then they've relied on HFS+. Use the disk format that suits you, TM will use the format that suits it.
