Will there be a large supply of 2011 iMacs?

There should be plenty bc they were shipped via boat rather than air. I'm guessing that they waited till they had many containers full rather than the small quantities they would put on an Airplane.
Also, with the media complaining about iPad 2 shortages, it wouldn't be smart for Apple to not have enough iMacs. Although demand is obviously different, it's in Apple's best interest to have enough at launch.

If you know for sure you are going to buy one then go down to your Apple store and see if you can reserve one.

I am not sure about when things are first released though because I remember when I went to the Apple store when the 27" i7 Quads were first released they didn't have the high ends in stock yet, they were only available online..
I called up the Apple Store 2 days ago as I was about to buy a high end iMac (just before I found out about the upcoming refresh).. I asked if only custom specs were available through the online service and they said that they can do it all in store
This is not true at all in my experience. They can do minor upgrades like more main memory, but if you want a custom HDD/SSD setup, they cannot do that for you in-store.
Oh ok sorry, you are probably right as I did not ask for the SSD option I only asked for the i7 and 8GB RAM
This is not true. BTO models can only be configured and purchased online. Some stores may stock already-configured BTO models, but you can't reconfigure them in the store.
The problem is that they often do not know. They don't get advanced warning about what they are getting shipped. Whatever shows up on the dock that morning is what they get. So they can say that they probably will be getting stock, but they might not know for sure.
I'd suggest calling them right after the release announcement and asking what they've got in stock.
Exactly! Which is why no one else knows, either. Personally, if I were ready to buy, I'd avoid all the release-day mayhem and let things die down first. I also only buy what I need, when I need it from available models, so I don't worry about upcoming releases. Anything you buy will be an "older model" within a year or so, so it's pointless to try to stay on the "bleeding edge" of technology updates. But that's just me.

In my experience, the only thing they can upgrade in the store is memory. But the cost of memory purchased from Apple is much higher than getting it from Crucial or other suppliers. Larger HDD or SSD and more powerful processor options have to be ordered BTO
