1. Turn-by-turn - I just went on a long vacation in my car and my dedicated Garmin unit was a trip saver. With the right software the iPhone should have the same functionality.
2. Topographical maps - I've used (and created) topo maps for hiking. It would be great to have this functionality integrated into the iPhone, too.

The signal-dependant streaming maps that many users on this site associate with "GPS" is an insult to the real deal. It has its own uses but is not a true substitute.
 
OK, so someone revives this thread after 2 years, and now why is it still going on?! The iPhone has had GPS since June 2008, yet people TODAY said why it should?
