How do you get Tweetbot alpha to show up in Notification Center?

When you first log in it should ask you to allow notifications.

How do you get Tweetbot alpha to show up in Notification Center?

Go to the Tweetbot's preferences and you'll see Notifications tab on top. You do need to make sure to update Tweetbot, notifications was added in Alpha 2, they're at Alpha 3 now.
