Any luck if you try to do it from System Preferences under Display?

It seems to be working fine for me on the same beta.

On Sierra 10.12.3 all works perfectly.

I was wondering if you found a fix for this. I just updated to 10.12.4 (final) and I got the same issue...

Nope. Yesterday I spoke with Apple Support. They recommend me fresh install. That's all Today I created a back up and after work try to fresh install. I reply here if this helped me.

So guys one more moment... I did fresh install 10.12.3 and brightness controller work perfect, after I download update to 10.12.4 MAS. So, what did you think? Right - brightness broke. Let's try fresh install of 10.12.4 eta 40 minutes. I really mad now.
[doublepost=1490824449][/doublepost]So. Fresh install 10.12.4 - brightness not work. Now I even don't have a word.

Update: I spoke with Apple Support one more time. Capture and send them all data and screens... SO
"It appears to be a software issue, with the way we isolated it, if it's a bug it's something that could be fixed in an update but there's still a chance of a setting causing it that can be changed.

Yes, I will keep in contact, I will be the first ot know of changes or responses from engineering. Apologies for the response time, i'm writing up all the details, every little thing you tried and tested has to be detailed exactly."

For now use 10.12.3. That's all guys. Stay frosty.

Sad to hear it :(

I found temporary solution for 10.12.4 — Brightness Slider from app store works for me

@idoo if you click on the link and read what it is referencing it will become clear. Hint: the bug seems to center around the Ambient Light Sensor and its settings.
