If I buy 2 16GB RAM cards to fill the other 2 slots, will these work along with the two 4GB that come stock for a total of 40GB RAM? Or do they all need to be the same size in each slot?

Yes, they will. That would be my plan. Order stock ram (2x4GB) from Apple, get 2x16GB and install myself for 40GB total. I've been using 16GB at home in a PC, and 20 and 32 in 2 different iMacs at work with no problem, so 40 would be a dream!

And if for some reason you find yourself exceeding the limits of 40GB of ram, buy another 2x16 in the future and sell your original 2x4GB on eBay for a small bit of cash.

My choice will be end 27". But, 512 SSD or i7? I have money only for one of them. I am not hard user, but I want to keep my Macs for many many years.
 
I ordered the Low end 27" with 1TB SSD upgrade, extra trackpad and expanded keyboard, should be sufficient for the next 5 years of photo editing, netbrowsing and kids stuff I hope:
