But I did not understand this, if I am playing movie through iTunes why would anyone want to use Apple TV in it ? Where does Apple TV fot in this entire scenario? How will it help and what will it do ? Or rather what can it do ?
Assuming you have :
a) an Apple TV connected to your big TV / home cinema and
b) a Mac with Home Movies / Films in your local iTunes (library)
Then you can simply start a movie on your Mac / iTunes and (wirelessly) send it to your big TV without any Cloud subscriptions or streaming plans and without configuring any network sharing.

Only exception I have is, a windows computer and the video file is saved on My Windows Computer Desktop ? Anything that I can leverage here ?

That too I want Audio streaming from Airplay-1 speakers and Homepod. Possible ?

It doesn't matter if it's a Windows iTunes, it still works exactly as described (just tested with a Windows 10 machine).

I'm not 100% sure I understand the question, but here's what I observed:
- Sending music to several different Airplay speakers at once with (Windows) iTunes works
- When playing a video in (Windows) iTunes, Airplay is only possible to one receiver at once and it has to support audio AND video (like the Apple TV).
Maybe 3rd party software for Windows exists that achieves what you seek but I am unfamiliar with any.

This deviates from OPs original question, but if watching personal/local movies on a TV is a regular thing, I'd also suggest Plex.
Plex Media Server on the Windows PC, movie files locally on said PC or a NAS etc. and on the Apple TV or Smart TV the Plex App.
For a quick solution without setting up Plex, instead of Airplaying the movie from iTunes to the Apple TV, enable Home Sharing in iTunes and simply access your media from the Apple TV via the integrated TV app.

Let me be sure if I understand you correctly. You are saying have the Local File uploaded on iTunes and then enable home sharing in iTunes and then through the TV app access it on iTunes ? is this possible ? Never knew or heard this could be done ?

Yes, it‘s a well documented feature (and working fine). Local video files in Windows iTunes, browsing and playback of said files through TV app on the Apple TV.

When playing music, none of your Airplay (1 or 2) speakers show up in iTunes? If they are all in the same network, only a firewall issue comes to mind.
Or do you mean videos? As previously said, for videos in Windows iTunes, you can’t select plain Airplay speakers as target.

Yes, I mean when playing Videos in iTunes.I am not getting even my Homepods OG (Airplay-2) show in iTunes to Stream Audio to them ?
